Off-label Use For Treatment Resitant Depression

Clinical trials were looking at the use of opiod agonists as off label anti-depressants. Since they block the release of glutimate without the more addictive opiate side effects. Also, releases more dopamine? Ketamine also being studied as well as anti-inflammatory drugs in combination with opioid agonists? I am probably mistating the complex bio-chemistry involved, but bottom line it seems like anti-depressant meds are all moving toward reducing inflammation by reducing glutamate release, (or is it the other way around) from the spinal column by binding opioid agonists to the pain receptors. Thereby also increasing dopamine release. I know several experimental drugs were in the pipeline but some fizzled out. Any ideas as to truly "new" anti-depressants for treatment resistant major depressive disorder? Bupronorphine hydrochloride was also mentioned frequently.

RE: "Any ideas as to truly "new" anti-depressants for treatment-resistant major depressive disorder?"

In my opinion, the idea of something being "treatment-resistant" is clinically biased for each individual's unique case of depression (or whatever condition it is that ails them). That said, to each their own and what works well for some people may not work for others. I think if we were able to come up with a long list of things we've all tried for depression, perhaps others can use it as a template for working with their doctor or in some cases self-medicate (responsibly) with alternative non-pharmaceutical options.

I do believe that opioids, when administered responsibly, can help alleviate depression - especially if it stems from pain or other related bodily discomforts. However, in today's opioid climate, this is but a dream and I feel that Kratom is a wonderful alternative.
