Have you discussed this with your doctor?
How long have you been taking them, at the current dose?
The Cholchicine has really only been shown to be effective a treating gout, however, the Tramadol is an analgesic used to treat pain.
But there is always the possibility that these just aren't the right medications to help with your pain, or if you've been taking them for a long period of time, you may have grown tolerant to them and require a dosage adjustment.
