They can both increase serotonin levels, which could cause serotonin syndrome. However, the doesn't mean they can't be taken together, if your doctor has prescribed them both, it just means that they have to carefully calculate the dosage and they should be monitoring you regularly.
The FDA classifies Tramadol as an opiate, so it has the potential to be habit forming and may cause side effects, such as nausea, dizziness, drowsiness, dry mouth and constipation.
The side effects for Effexor may possibly include nausea, dizziness, headache, dry mouth, mood swings and weight changes.
