If I take an OxyContin 80 mg at 10 PM Monday night, and on Tuesday morning I take one 80 mg + one 30 mg Oxycodone at 7 AM would it be working in my body by 8:30 that morning? Thanks

Yes, based on your dosage schedule I'd say it would definitely be working in your body by then, regardless of whether you take immediate release or a delayed release formulation. For instance, conventional oxycodone has a peak plasma concentration in your blood stream of about 1 hour after taking it, whereas the time-released version would reach its peak after about 3 hours. Half life can be up to about 4 hours, and traces of it remain in your body for up to 72 hours (but by that time you would probably have taken your next dose if you are on a regular pain management schedule).
