It is true that they cannot take action against legally prescribed medications.
However, it seems you are worrying needlessly. Under right to work, an employed can fire any employee, at any time, for any reason or for no reason. Therefore, if they really want to get rid of someone, they will do and there is nothing that can be done about it.
But, if you refuse the test, you are definitely giving them grounds.
