Mounjaro And Liver Markers

I was doing so well with Mounjaro, but my Doctor took me off it because of significantly increased liver enzymes / markers in a routine blood test. I am so disappointed and have been doing as much on line research as I can. So surprising given that reduced liver fat is supposed to be one of the benefits. I have read that it could be the speed of weight loss, rather than the Mounjaro itself, that caused this. Can anyone relate?

I am pretty sure Mounjaro can induce (increase its activity) while the drug maker is trying to hide the fact. And may affect other enzymes. Although the relationship may be complex and could potentially go either way with enzyme activity.

Forskolin increases intracellular cAMP, which affects some P 450 enzymes like CYP3A4. If you look deeper into the GLP-1 related drugs research, their primary activity is by GLP-1, and in the case of Mounjaoro added GIP, increasing intracellular cAMP even better than GLP-1 alone. Which activates kinases and does all those things we think of like delayed gastric emptying, enhanced insulin release and attenuating insulin resistance. Which Forskolin also does but is far weaker at it. But, Forskolin is considered a "potent inducer" of CYP3A4 in some research. Elevating intracellular cAMP also affects some P 450 enzymes and CYP3A4 in particular. Which when elevated can cause liver issues. Especially if someone takes acetaminophen frequently.

N-Acetyl-Cysteine (NAC) is given to people to protect their liver from damage from acetaminophen overdose. Which involves P 450 enzymes. So, supplementing "may" help protect your liver, even if not taking acetaminophen.

Choline has been shown to be liver protective in clinical trials. RDA of choline is 550mg. Which many people may be short on especially with reduced food intake.

Taurine has mixed results, so "may" be protective also.

I'm on Mounjaro and have several drugs and supplements messing with my CYP3A4 enzyme with the net result of it being elevated. I do take 2 grams taurine in the morning and 1 in the evening. I take choline supplements a few times with meals during the day (alpha GPC, choline bitartrate). and 1,200 mg NAC in the morning and evening. My liver blood tests are all good. Not proof of anything as a single data point with no control group or screening of other factors. But, "may" indicate worth while looking into more.

Also note, good to take vitamin B-complex in the morning with NAC as it prevents elevated homocysteine levels if deficient in some B vitamins. Plus B vitamins have also been shown to be liver protective. All the supplements mentioned are shown to have some benefits although research is lacking full scale large trials for most of them. NAC is FDA approved to protect the liver in acetaminophen poisoning so has more science and clinical research behind it.

I did see that Mounjaro maker had done some test tube tests with human liver cells and Mounjaro and concluded it had no P 450 activity. My unprofessional opinion, Mounjaro works by gradually increasing cAMP levels. Claiming no effect based on short term tests for a drug that can take weeks or months to reach its full effect is dishonest at best. Forskolin is acute and increases cAMP activity with each dose. Which is why they see it affecting liver enzymes and why I am skeptical of Eli Lilly in that regard (among others not listed here).

I am a huge advocate of GLP-1 drugs with the caveat that they are hiding a few things and people are paying the price from the lack of full disclosures while they rake in billions.
